Simple question...

When a label says "Calories 240 - Calories from fat 40"

Does that mean It has 240 calories, and 40 of them are from fat, Or does that mean my nice little cup of yogurt actually has 280 calories?

nope it's got 240 calories it's just showing you the amount of those calories that are from fat (many people try to keep fat within the 30 percent range of total calories for the day so it helps you with the calculations0

I beleive 1 gram of fat has 9 calories - so you could get that number on your own -

correct. 1 gram of fat contains 9 calories. for a sound weightloss program, you should not consume more than 20% fat of your total calories. saturated fats should be cut to no more than 10%. these are daily values.
